Serotonin and Treatments for Depression, Animation.

According to the Journal of Advances in Psychopharmacology, author Penn advises that since there is little to no evidence available to assess the efficacy of SSRIs, they are generally considered working if a person can tolerate the side effects.15

If you feel your antidepressant medication is no longer working, or you are dissatisfied with the side effects of your current medicines, talk to your doctor. It may be helpful to contact a nutritional psychiatrist20 or other holistic practitioners familiar with alternative treatments for depression. It is of extra importance for your safety to have the proper support when changing medications or adjusting dosages.21 If your medication no longer works, there may be better alternatives. How Trintellix Works To Treat Depression

The FDA, drug researchers, and drugmakers all admit the precise action of how antidepressants work is unknown.4,12 There has been much research done to clarify the mechanisms of action and how these relate to mental illness, but so far nothing definitive has been established beyond some quite detailed theories and hypotheses, all as yet unproven according to published research in The Lancet.11

Abrupt Discontinuation Of Trintellix Treatment

Patients who are taking Trintellix with dosages of 10 mg, 15 mg, and 20 mg per day have been evaluated for discontinuation symptoms using a scale called Discontinuation-Emergent Signs and Symptoms in clinical trials.

In the first seven days of abrupt discontinuation of Trintellix with dosages of 15 mg/day and 20 mg/day, there were patients who experienced the following discontinuation symptoms:

  • Mood swings
  • Muscle tension
  • Sudden outbursts of anger

There should be appropriate monitoring and close observance of all patients who are under antidepressant treatment for any indication. The following symptoms must be closely monitored:

  • Suicidal tendencies
  • Worsening of depression
  • Unusual behavioral changes during the first few months of treatment or when increasing or decreasing the drug’s dosage

When changing a patient’s therapeutic regimen, consideration should also be given with regards to the following:

  • The possibility of discontinuing the medication
  • A persistently worsening depression
  • The emergence of suicidal ideation
  • Abrupt and severe symptoms before the occurrence of a worsening suicidality or depression
  • The onset of certain symptoms that were not initially experienced by patients

Vortioxetine Treatment Of Depression In Parkinsons Disease

M. Russo, C. Carrarini, F. Dono, L. Ferri, M. Di Pietro, M. Onofrj

Session Time: 1:45pm-3:15pm

Location: Agora 3 West, Level 3

Objective: To assess efficacy and safety of Vortioxetine in Parkinsons Disease patients affected by depression and its effects on motor performance.

Background: Depression is one of the most common non-motor symptoms in PD patients, with a reported prevalence of 3040% overall. It is associated with rapid progression of motor and cognitive symptoms, as well as with worse quality of life. Vortioxetine is an antidepressant drug, acting as multimodal regulator of serotonin trasmission. Several types of antidepressants are currently available to treat PD-associated depression . Nevertheless, no previous studies were carried out to assess the effects of Vortioxetine in dPD.

Results: In this cohort, mean baseline scores were: MDS-UPDRS 19±5 HDRS 18±6. After three months of 10 mg daily Vortioxetine treatment, patients showed improvement of HDRS score , as well as patients treated with 20 mg dose . MDS-UPDRS scores were not significantly modified by the therapy in both groups. No severe side effects were reported.

Our findings suggest that Vortioxetine improves depression severity in dPD patients, without significant worsening of motor symptoms. Moreover, Vortioxetine was proved to have a remarkable safety profile.

Movement Disturbances Associated With Ssris

Raphael J. Leo, MDPsychiatric Times

The SSRIs have become the most widely prescribed antidepressants in the United States. With this increased use has come more information on adverse events associated with their use, such as sexual dysfunction. However, movement disturbances associated with SSRI use can adversely impact treatment. This article explains the pathology of movement disorders and describes confounding variables such as other medications, pre-existing neurological insults and the nature of case reviews. Those at risk include the elderly, patients taking neuroleptics and those exposed to high levels of SSRIs.

The selective serotonin reuptake inhibitors are employed to treat an array of disorders, including depression, social phobia, panic disorder, posttraumatic stress disorder and obsessive-compulsive disorder. With generally fewer side effects and better tolerability than tricyclic antidepressants and monoamine oxidase inhibitors, the SSRIs have become the most widely prescribed antidepressants in the United States.

In a review of the literature, 71 cases of SSRI-associated extrapyramidal symptoms were found . Akathisia was most common, followed by dystonia, parkinsonism and tardive dyskinesia-like states. In addition, 16 cases of worsening parkinsonism were found in patients with pre-existing Parkinson’s disease.

Vortioxetine Ameliorates Motor And Cognitive Impairments In The Rotenone

Non-dopaminergic injury leads to non-motor symptoms in PD.

Vortioxetine attenuates rotenone-induced motor and cognitive impairments.

Vortioxetine may cause a mild dysfunction in SNpc tissue.

Vortioxetine alters neurotransmitter and cytokine levels in PD rat brains.

Targeting TLR-2 signaling may help to fight motor and non-motor symptoms in PD.

Trintellix And Herbs And Supplements

Trintellix can interact with certain supplements or herbs you may be taking. Supplements that affect serotonin levels can increase your risk of developing serotonin syndrome.

Examples of these supplements include:

  • 5-HTP

Trintellix is a unique antidepressant medication because of the way it works. Its sometimes called an atypical antidepressant or a multimodal antidepressant.

Depression may be partly caused by decreased levels of brain chemicals such as serotonin.

Like many antidepressants, Trintellix increases serotonin activity by blocking the reabsorption of serotonin in the brain. This increase in serotonin might help balance out levels of brain chemicals to help improve symptoms of depression.

Unlike other antidepressants, Trintellix also binds to and blocks certain types of serotonin receptors in the brain. Receptors are like docking stations on cells, and serotonin attaches to them.

When Trintellix blocks these receptors, serotonin cant attach to them. Its not clear how this action helps relieve depression.

Trintellix Vs Effexor Xr

Trintellix and Effexor XR are both prescription antidepressant medications. They both affect serotonin, a chemical messenger in the brain, but Effexor XR also affects norepinephrine.

Uses

Trintellix and Effexor XR are both FDA-approved for treating major depressive disorder in adults. Effexor XR is also approved for other uses, including:

boxed warning regarding the risk of suicidal thoughts and behaviors in certain people. This is the most serious warning from the Food and Drug Administration . Boxed warnings alert doctors and patients about drug effects that may be dangerous.

Effectiveness

The only condition both Trintellix and Effexor XR are used to treat is MDD. The effectiveness of these drugs in treating MDD hasnt been directly compared in clinical studies. However, an showed that Effexor XR may be slightly more effective than Trintellix.

Cost

Trintellix is only available as a brand-name drug. Effexor XR capsules are available as a brand-name drug and in a generic form called venlafaxine.

Effexor XR is similar in price to Trintellix. Generic Effexor XR is typically less expensive than both brand-name products. In any case, the actual amount you pay will depend on your insurance plan.
