Analysis Of Fatty Acids By Gc/ms
The fatty acid composition of lipids in all tissues was analyzed by GC/MS after conversion to methyl esters. Briefly, the lipids were evaporated under N2 and dissolved in 0.5ml toluene containing 25g of 22:3 free fatty acid and 250g butylated hydroxytoluene. Methanolic HCl was then added, and the reaction mixture was heated under nitrogen at 100°C for 1h. The acid was neutralized by adding 1.0ml of 0.33N NaOH, and the fatty acid methyl esters were extracted twice with 3ml of hexane. The pooled hexane extracts were evaporated under nitrogen and re-dissolved in 30µl of hexane and 1µl was injected into GC/MS. The analysis was carried out using a Shimadzu QP2010SE GC/MS equipped with a Supelco Omegawax column as described previously. Total ion current in the range of 50400m/z was used to quantify the fatty acids, using 17:0 as the internal standard.
